Abstract— The objective of the present experiments was to correlate changes in cellular energy metabolism, dissipative ion fluxes, and lipolysis during the first 90 s of ischemia and, hence, to establish whether phospholipase A2or phospholipase C is responsible for the early accumulation of phospholipid hydrolysis products. Ischemia was induced for 15–90 s in rats, extracellular K+ (K+e) was recorded, and neocortex was frozen in situ for measurements of labile tissue metabolites, free fatty acids, and diacylglycerides. Ischemia of 15-and 30-s duration gave rise to a decrease in phosphocreatine concentration and a decline in the ATP/free ADP ratio. Although these changes were accompanied by an activation of K+ conductances, there were no changes in free fatty acids until after 60s, when free arachidonic acid accumulated. An increase in other free fatty acids and in total diacylglyceride content did not occur until after anoxic depolarization. The results demonstrate that the early functional changes, such as activation of K+ conductances, are unrelated to changes in lipids or lipid mediators. They furthermore suggest that the initial lipolysis occurs via both phospholipase A2 and phospholipase C, which are activated when membrane depolarization leads to influx of calcium into cells.  相似文献

A new chemolithotrophic bacterial metabolism was discovered in anaerobic marine enrichment cultures. Cultures in defined medium with elemental sulfur (S0) and amorphous ferric hydroxide (FeOOH) as sole substrates showed intense formation of sulfate. Furthermore, precipitation of ferrous sulfide and pyrite was observed. The transformations were accompanied by growth of slightly curved, rod-shaped bacteria. The quantification of the products revealed that S0 was microbially disproportionated to sulfate and sulfide, as follows: 4S0 + 4H2O → SO42- + 3H2S + 2H+. Subsequent chemical reactions between the formed sulfide and the added FeOOH led to the observed precipitation of iron sulfides. Sulfate and iron sulfides were also produced when FeOOH was replaced by FeCO3. Further enrichment with manganese oxide, MnO2, instead of FeOOH yielded stable cultures which formed sulfate during concomitant reduction of MnO2 to Mn2+. Growth of small rod-shaped bacteria was observed. When incubated without MnO2, the culture did not grow but produced small amounts of SO42- and H2S at a ratio of 1:3, indicating again a disproportionation of S0. The observed microbial disproportionation of S0 only proceeds significantly in the presence of sulfide-scavenging agents such as iron and manganese compounds. The population density of bacteria capable of S0 disproportionation in the presence of FeOOH or MnO2 was high, > 104 cm-3 in coastal sediments. The metabolism offers an explanation for recent observations of anaerobic sulfide oxidation to sulfate in anoxic sediments.  相似文献

A sequential five-step extraction scheme for phosphorus pools in freshwater sediment was modified for use in marine sediments. In the second step phosphate bound to reducible forms of iron and manganese (iron-bound P) is extracted by a bicarbonate buffered dithionite solution (BD-reagent). The extraction scheme was tested on sediment from 16 m water depth in Aarhus Bay, DK and used in two other marine sediments: Kattegat at 56 m and Skagerrak at 695 m depth. By comparing the BD-extractable P-pool with both the pool of iron in the BD-fraction and the pool of oxidized, amorphous or poorly crystalline iron (am.FeOOH), highly significant correlations (p < 0.001) were observed in all three sediments. Thus, we conclude that the BD-reagent was very specific for iron-bound P. Further evidence for this came from two experiments: 1) Enhanced BD treatment did not result in additional phosphate extraction and 2) by sequential extraction of phosphorus pools in pure cultures of diatoms and cyanobacteria no phosphate was recovered in the BD-fraction. The pool of am.FeOOH was very important for controlling porewater phosphate concentration which was inferred from the significant inverse relationships between the two parameters (p < 0.001) in all sediments studied. Further, an isotopic exchange experiment with 32POf4/p3– revealed that BD-extractable P was by far the most exchangeable P-pool even deep in the sediment where the pool size was small. Iron-bound P made up 33–45% of total P in the surface sediments. The ratio between iron-bound phosphate and am.FeOOH was 8–11 in Aarhus Bay and Kattegat. In Skagerrak the ratio was 17, which may indicate that the iron mineral extracted from this sediment is less capable of adsorbing phosphate or less saturated with phosphate.  相似文献

-Chymotrypsin was modified with cyanuric chloride activated monomethoxypolyethylene glycol (MPEG) with molecular weights 1900 and 5000. Using the higher molecular weight MPEG a product that was soluble in benzene at moderate levels of modification was obtained, whereas with MPEG 1900 almost all the enzyme's amino groups had to be modified for dissolving the conjugate. The catalytic activity decreased with increasing degree of substitution. Apparent Vmax was considerably higher for the less modified enzyme preparation than for the more modified one, while Km,app stayed almost constant. The modified enzyme was used for peptide synthesis. The reaction was dependent on the content of dissolved water. Both Vmax,app and Km,app increased with increasing water content. It was possible to achieve a process with complete conversion of substrate to dipeptide.  相似文献

岭南垛基果林湿地是珠三角地区典型的湿地类型之一,其对土壤碳汇的贡献值得关注。为探讨果林种植类型对土壤有机碳的影响,对广州垛基果林湿地内种植黄皮(Clausenalansium)(HP),龙眼(Dimocarpuslongan)(LY)、杨桃(Averrhoa carambola)(YT),龙眼和黄皮间种(LH),杨桃、龙眼和黄皮间种(YLH)共5种种植类型下的表层(0~20 cm)土壤碳组分进行研究。结果表明,不同的植被类型对土壤的总有机碳(SOC)、可溶性有机碳(DOC)、微生物生物量碳(MBC)、易氧化有机碳(ROC)、惰性碳(NLC)含量都有影响,LY的SOC含量最高(22.6 g/kg),显著高于YLH (P<0.05),且NLC含量显著高于LH和YLH (P<0.05)。NLC含量与土壤养分呈正相关,与土壤容重呈负相关。YT的MBC含量显著高于LY、HP、LH (P<0.05),且MBC/SOC显著高于HP、LY(P<0.05)。YLH模式下,土壤DOC含量和DOC/SOC显著高于其他植被类型(P<0.05)。LH的ROC/SOC显著高于HP和L...  相似文献

山医群体近交系中国地鼠的红细胞、白细胞、网织红细胞、血红蛋白及血清蛋白量均与人类相近。血小板数高于人类,但低于小鼠。嗜中性与淋巴细胞的比值与人类相反而与小鼠近似。血清尿素氮及谷丙转氨酶高于人类生理值,低于SD大鼠。心率为658±102次/min,呼吸率20±30次/min。心电图除T波低平外,其余与人类相近。颈动脉血压为13.47±1.01/12.48±1.09kPa。染色体数为2n=22。以上各项在近交系地鼠和野生地鼠间未见明显差异。  相似文献

在16℃、19℃、22℃、25℃.28℃,31℃和34℃的范围内, 草地螟(Loxostege sticticalis L.)成虫的产卵前期与温度的关系呈逻辑靳谛曲线,产卵雌蛾数、产卵历期和产卵量均以22℃为最高。初步确定19-25℃为草地螟蛾产卵的适温范围,22℃为最适产卵温度,16℃和34℃分别为产卵温度的下限和上限。成虫寿命与温度的关系为负相关(r=-0.91,P<0.01)。表明在试验温度范围内,温度越高,成虫寿命越短。但温度对雌雄成虫寿命的影响不一致,在28℃以下的温度内,雄蛾寿命比雌蛾长;在31-34℃的范围内,雄蛾寿命比雌蛾短。根据以上研究结果,对我国草地螟发生为害区的种群动态规律进行了讨论。  相似文献

聚谷氨酸(polyglutamic acid, PGA)作为一种天然多功能的聚合物,近年来成为研究的热点。由于很难通过化学方法合成,微生物发酵是目前生产聚谷氨酸的有效途径。【目的】从基因水平探究枯草芽孢杆菌聚谷氨酸合成途径中degS、degQ、degU、swrA、rocA、putM基因的功能,通过分子改造实现对代谢途径的调控。【方法】以枯草芽孢杆菌为出发菌株,通过对代谢途径中相关基因进行敲除或过表达,分别构建degS、degQ和degU基因缺失的重组菌,swrA、rocA和putM基因过表达的重组菌,借助菌株胞外聚谷氨酸积累的变化分析影响途径的关键节点。【结果】在摇瓶发酵条件下,重组菌Bacillus subtilis 168-swrA、Bacillus subtilis 168-rocA、Bacillus subtilis 168-putM的胞外聚谷氨酸含量分别是原始菌株的1.28倍、1.47倍和1.37倍。重组菌Bacillus subtilis 168-ΔdegS、Bacillus subtilis 168-ΔdegQ、Bacillus subtilis 168-ΔdegU的胞外...  相似文献

相比于氨氮,天然水体中的硝酸盐氮通常更稳定,导致更难将其从水中去除。由于好氧反硝化可以在有氧环境下进行反硝化作用去除硝酸盐氮,该过程对含有较高溶解氧的天然水体中硝酸盐氮处理有重要作用。本文综述了好氧反硝化菌的分离纯化现状、微生物代谢机制和环境影响因子,并介绍了功能菌群在微污染饮用水源水生物修复的应用研究进展。与一般的厌氧反硝化类似,好氧反硝化菌的种属分布较广,常见的如假单胞菌属(Pseudomoas)、产碱杆菌属(Alcaligenes)、副球菌属(Paracoccus)和芽孢杆菌属(Bacillus)等所属部分微生物均有好氧反硝化能力。大部分好氧反硝化菌株在最佳生长条件下(25–37℃、溶解氧浓度为3–5mg/L、pH为7–8、碳氮比为5–10)具有高效的脱氮效率。但目前好氧反硝化作用在微污染饮用水源水的生物修复方面的应用仍有着脱氮性能不稳定、菌剂流失等不足。此外,目前较少相关中试及实际工程应用的研究,需要进一步的深入探究。  相似文献

【目的】探究不同腐殖酸浓度下参与含砷水铁矿转化的微生物类群组成和丰度变化及对砷释放的影响,预测原位高砷含水层中功能微生物群参与有机质—含砷铁矿物转化过程对砷转化释放的作用。【方法】对河套平原高砷地下水和同深度高砷沉积物中的铁还原功能群落进行富集培养,构建室内厌氧微宇宙体系,将富集菌群分别加入到实验室条件下合成的不同浓度腐殖酸(0、1.5、7、14 mg C/L)-含砷水铁矿体系中,通过体系中砷、铁形态及浓度的变化分析,结合高通量测序技术、X-射线衍射(X-ray diffractometer, XRD),探究不同条件下砷的释放固定和群落的演替。【结果】高砷地下水组(G组)和沉积物组(S组)富集得到的铁还原功能群落具有明显差异,G组中以Aeromonadaceae为特殊优势菌群,而S组中以Shewanellaceae为特殊优势菌群。微宇宙实验结果显示,S组的铁还原量相对较高且速率较快;G组与S组中液相砷形态存在明显差异,整个培养期内G组均以As(Ⅴ)为主,而S组中前期以As(Ⅴ)为主,当反应到达20 d时液相As(Ⅲ)高达3.4μmol/L,推测此时具有砷还原功能的群落占优势地位。当反应...  相似文献
